چکیده

The levels of pyridine nucleotides in tissues have been the subject of a large number of reports. Much interest has been focused on the ratios of oxidized to reduced coenzymes in different pathological conditions (1). Recently, Glock and McLean (2) have reported the important observation that reduced TPN is present in whole rat liver in comparatively large amounts, whereas the concentration of the oxidized form of TPN was found to be very low.’ On the other hand, oxidized DPN was found to be present in an excess over the reduced form. Sensitive fluorometric procedures have recently been developed for the assay of different forms of the pyridine nucleotides (3). This has made possible an analysis of the subcellular tissue fractions. The present communication deals with the concentrations of DPN, DPNH, TPN, and TPNH in various cellular fractions and the factors which influence the levels of pyridine coenzymes in these fractions. This report will show that the most of the TPNH in rat liver is localized in the mitochondria.

N-demethylation of p-chloro-N-methylaniline catalyzed by subcellular fractions from the avocado pear (Persea americana).

Subcellular fractions from the avocado pear ( Persea americana) catalyzed formation of p-chloroaniline from p-chloro-N-methylaniline. Fractions prepared by centrifugation of avocado homogenates at 20, 000g for 20 min formed p-chloroaniline (2900 +/- 500 pmol min-1 mg protein-1) with an NADPH-generating system. p-Chloroaniline formation required reduced pyridine nucleotide (NADPH was 6-7 times m...

Dynamics of alaninaminotransferase activity in subcellular fractions of different areas of brain cortex and hypothalamus in postnatal ontogenesis under protein-free feeding regime and after its withdrawal

Total and specific activities of alaninaminotransferase (Al-AT) were determined in general tissues, mitochondrial and cytosol fractions of visual, orbital, motor, limbic areas of brain cortex and hypothalamus of three-month old and one-year old rats under 10-20 days and 30 days protein deprivation and under recovery of normal food regime during the same terms. It was found out that Al-AT activi...

Betaine accumulation and betaine-aldehyde dehydrogenase in spinach leaves.

Spinach leaf discs accumulated betaine when exposed to a mannitol solution of -20 bars. The accumulation was 12 micromoles per gram original fresh weight in a 24-hour period.Betaine-aldehyde dehydrogenase (EC 1.2.1.8) was assayed in various subcellular fractions prepared from spinach leaves, and it was found only in the soluble fraction. This cytosolic enzyme was purified 175-fold, and its prop...

Conversion and Distribution of Cobalamin in Euglena gracilis z, with Special Reference to Its Location and Probable Function within Chloroplasts.

Cobalamin is essentially required for growth by Euglena gracilis and shown to be converted to coenzyme forms promptly after feeding cyanocobalamin. Concentrations of coenzymes, methylcobalamin, and 5'-deoxyadenosylcobalamin, reached about 1 femtomole/10(6) cells 2 hours after feeding cyanocobalamin to cobalamin-limited cells. Cobalamins all were bound to proteins in Euglena cells and located in...
